Yes, but they must have separate node directories (tahoe -d <dir> ...), so that they can have separate identities and state.

The port you connect to is for the gateway/client node's web user interface. That needs to be unique, but you only need one.
Tahoe nodes connecting to each other automatically assign themselves ports for that purpose, unless you configure them to use a specific port, which you don't need to do unless you're trying to run a public server behind NAT port forwarding.
